  Our previous studies showed that cysteinyl leukotriene receptor 1 (CysLT1 receptor) antagonist pranlukast has a neuroprotective effect on global cerebral ischemia in rats.However, whether the protective effect of pranlukast is its special action or a common action of CysLT1 receptor antagonists remains to be clarified.This study was performed to determine the change of CysLT1 receptor expression in global ischemic injury and to evaluate the effect of montelukast, another CysLT1 receptor selective antagonist, on hippocampal neuronal damage after global ischemia in rats.Global ischemia was induced by bilateral common carotid artery occlusion combined with hypotension in rats.CysLT1 receptor expression was detected by immunohistochemistry.Montelukast was orally administered.The number of survival of CA1 neurons was assessed.CysLT1 receptor expression was up-regulated in the ischemic hippocampus after global cerebral ischemia.Treatment with multiple doses of CysLT1 receptor antagonist montelukast (0.5 and 1.0 mg/kg, before and after ischemia)significantly attenuated CA1 neuronal damage in a dose-dependent manner.Furthermore,protective effect of montelukast (0.5 mg/kg, 5 consecutive days) on hippocampal neuronal damage was still evident 30 days after ischemia, providing the experimental evident that montelukast confer a long lasting neuroprotection.CysLT1 receptor mediates hippocampal neuronal damage after global cerebral ischemia, CysLT1 receptor antagonist montelukast has a dose-dependent and a long-lasting cerebroprotective effects on hippocampal neuronal damage following global ischemia in rats.
