目的分析150例初、复治涂阳肺结核患者2个月末的痰标本进行Bact/ALERT 3D仪液体培养和固体罗氏培养情况。方法对治疗2个月后150例初、复治涂阳肺结核患者的2个月末的痰标本进行Bact/ALERT 3D仪液体培养,阳性培养物送浙江省疾控中心进行HAIN分子生物学试验,并与固体罗氏培养法进行比较。结果 150例中,涂阳37例,液体痰培养阳性29例;涂阴113例,液体痰培养阳性65例。菌型鉴定结果均为结核分枝杆菌。传统药敏试验结果,耐药19例(其中:广泛耐药2例,耐多药6例,单耐药8例,多耐药3例)。Bact/ALERT 3D液体快速培养法从接种到HAIN实验出结果,平均20.5 d;传统罗氏培养法时间平均3个月。Bact/ALERT 3D液体快速培养法未见污染,传统罗氏培养法污染7例。结论 Bact/ALERT 3D液体快速培养法检测结核分枝杆菌,具有阳性分离率高,能缩短报告时间,污染率低,安全可靠,自动化程度高,在耐药结核病早期诊断中具有应用价值。
Objective To analyze the liquid culture and solid Roche culture of sputum samples from 150 patients with initial and retreatment smear-positive pulmonary tuberculosis at the end of 2 months. Methods Bact / ALERT 3D liquid culture was performed on the sputum specimens of 150 patients with initial smear-positive smear-positive pulmonary tuberculosis after 2 months of treatment. The positive cultures were sent to Zhejiang CDC for HAIN molecular biology test. Compare with solid Roche culture method. Results In 150 cases, smear-positive in 37 cases, liquid sputum culture positive in 29 cases; smear-negative in 113 cases, liquid sputum culture positive in 65 cases. Mycobacterial identification results are Mycobacterium tuberculosis. Traditional drug susceptibility test results, 19 cases of drug resistance (including: 2 cases of extensive resistance, multidrug resistance in 6 cases, single drug resistance in 8 cases, multi-drug resistance in 3 cases). Bact / ALERT 3D liquid rapid culture method from inoculation to HAIN experimental results, an average of 20.5 d; traditional Roche culture method for an average of 3 months. Bact / ALERT 3D liquid rapid culture method no pollution, traditional Roche culture pollution in 7 cases. Conclusion Bact / ALERT 3D liquid rapid culture method for detection of Mycobacterium tuberculosis has high positive separation rate, shortened report time, low pollution rate, safe and reliable, high degree of automation, and is of value in the early diagnosis of drug-resistant TB.
