东方瓜馥木(Fissistigma tungfangense)是瓜馥木属(Fissistigma)植物,性温味辛,具有祛风除湿、活血止痛之功效,主治风湿痹痛、腰痛及跌打损伤,海南民间多用于治疗各种风湿痹痛、关节炎等[1,2]。本课题组对东方瓜馥木的乙醇提取物进行抗类风湿活性初步测试,结果显示该植物的提取物均有抗类风湿性关节炎作用。
苯基非那烯酮是香蕉的植保素,具有抗菌抗线虫活性[1,2],按结构可分为Ci9苯基非那烯、C18O氧杂苯基非那烯、C18N氮杂苯基非那烯酮[3].本研究发现C18N氮杂苯基非那烯酮类生物碱主要分布于血皮草科植物Xiphidium caeruleum花的雄蕊(图.1),并从中分离鉴定氮杂苯基非那烯酮类化合物19个[4].运用稳定同位素13C标记和组织培养技术,构建了该植物中苯基非那烯酮的生物合成及代谢

目的：寻找FK228生物合成基因簇dep中LysR家族转录调控蛋白DepR的结合位点.方法：将N端带有His标签的重组蛋白DepR在大肠杆菌中进行表达和纯化.PCR扩增dep基因簇中的基因间隔区域.通过凝胶阻滞实验确定重组蛋白DepR是否与其结合.结果：在dep基因簇中并没有发现DepR的结合区域，而基因簇下游与FK228生物合成无关的基因orf21(编码MinC同源蛋白，可能参与细胞分裂)的启动
