Aim: To discover novel PPARy modulator, PPARy-Gal4 chimera receptor and its mutant derivatives were applied to evaluate the binding activity of ligands and their molecular basis of interaction.Methods: To construct a series of chimera receptors, the plasmids of pcDNA3.1 Gal4 were inserted with PPARγ ligand binding domain fragment or the amino acid site mutant derivatives, such as Ser 273, Ser289, His 323, His 449, and Tyr473, and were cotransfected with a Gal4-promoter regulated luciferase reporter gene plasmid in 293E cell.
