The excessive generation of reactive oxygen species (ROS) is a primary event in cerebral ischemia/reperfusion (I/R), contributing to brain injury.In the present study, we studied the neuroprotective effects ofPanax notogineng extracts against cerebral I/R in hippocampus.Panax notogineng extracts were injected intraperitoneally twice for 2 days and then transient mouse middle cerebral artery occlusion (MCAO) model of brain I/R was performed.2,3,5-triphenyltetrazolium chloride (TTC) staining was used to indicate infarction volume.Our results showed that Panax notogineng extracts markedly decreased infarction volume induced by MCAO.As well as Panax notogineng extracts significantly improved neurological behaviors by using Longa scoring method.Panax notogineng extracts decreased malondialdehyde (MDA) concentration in hippocampus.The decrease ofpro-caspase 3 and Copper/zinc-superoxide dismutase (SOD-1) expression in MCAO groups were restored by treatment with Panax notogineng extracts.Notably, expression of heat shock protein 70 (HSP70) were significantly higher in MCAO groups but their expression were more significant in Panax notogineng extracts treated groups.Panax notogineng extracts significantly restored the levels of Akt which were severely reduced in MCAO groups.And Panax notogineng extracts treatment significantly activated nuclear factor-kappa B (NF-κB).These findings demonstrate that Panax notogineng extracts exert the neuroprotective effects by ameliorating redox status and activating anti-apoptotic signal pathway.
