α-Arbutin (4-hydroquinone-α-D-glucopyranoside) is a powerful skin-whitening agent that blocks melanin biosynthesis by inhibiting tyrosinase activity, and its biological activity is 10-fold higher than that of β-Arbutin (4-hydroquinone-β-D-glucopyranoside).Therefore, α-arbutin is widely used as a cosmetic and pharmaceutical.Here we described the development of a biocatalyst for efficient and rapid production of α-arbutin.We used Escherichia coli that expresses recombinant amylosucrase from Xanthomonas campestris (XcAS).The specific activity of recombinant XcAS was (specificity activity 40.54 U mg-1).After optimization, 1 M sucrose and 200 mM HQ were used to produce 53.86 g L-1 of α-arbutin.The prodcutivity of α-arbutin reached 26.93 g L-1 h-1, which, to the best of our knowledge, is the highest reported.The structure of α-arbutin was confirmed using 1H and 13C nuclear magnetic resonance.The E.coli whole-cell biocatalyst that expresses XcAS offers a simple and inexpensive method for efficient and rapid production of α-arbutin that shows great promise for use by the pharmaceutical and cosmetics industries.
