In this paper,sterols in apple seed oil were separated and quantitatively analyzed by thin layer chromatography (TLC) and P-S-Te spectrophotometry respectively.The composition of developing solvent and the condition for color reaction were studied,and the precision,accuracy and stability of this method were discussed as well.The result showed the optimum composition of developing solvent for TLC was ether:petroleum ether=3∶7(V/V).After TLC separation,a purple band was observed under ultraviolet lamp.When it reacted with P-S-Te reagent,stable green matter was formed,which has the maximum absorption at the wave length of 660nm.The optimum conditions of color reaction were:ethanol 2mL,P-S-Te reagent 2mL,temperature 50℃,time 15min.This analysis method has many advantages,such as fast,with high precision and good reproducibility,easy to operate etc.
