Until now, bovine embryonic stem cells (bESCs) line with stable pluripotency in vitro has not been established.Different from mouse, a key trophectoderm (TE) regulator, Caudal-related homeodomain factor CDX2 expressed in TE and inner cell mass (ICM) in bovine blastocysts.Therefore, we hypothesized that expression of CDX2 in bovine ICM may be a barrier to pluripotenct maintenance of bESCs.In this study, CDX2-deficient bovine embryos were produced by SCNT.Expression and localization of pluripotent genes in CDX2-deficient bovine blastocysts were not affected.Bovine embryonic stem-like cells (bESLCs) were successfully derived from these embryos, revealed more robust pluripotency after a long-term cultivation.
