Autophagic pathway is essential for the neuroprotective effect of T10 in PD model

  Parkinsons disease (PD) is a neurodegenerative disease that results from the progressive loss of dopaminergic neurons in the substantia nigra of the midbrain.Recently, dysregulation of the autophagy pathway has been observed in the brains of PD patients and in animal models of PD, indicating the emerging role of autophagy in this disease.Triptolide T10 is a monomeric compound isolated from Tripterygium wilfordii Hook.f, a traditional Chinese herb for anti-inflammatory medications.It has been reported that T10 induced cell death by autophagic pathway in tumor cell line and disrupted autophagy leads to dopaminergic axon and dendrite degeneration.In our study, we found that T10 protected MN9D cells against MPP+ injury.To better understand the mechanism of T10, we investigated the autophagic pathway in this process and found T10 could increase in the LC3 punctate staining pattern and autophagy-specific LC3-Ⅱ protein expression in a time and dose dependent manner compared with untreated cells.These results indicated that autophagic pathway might play an essential role in T1 0s neuroprotective effect.
