In the present study,a fast method using high performance liquid chromatography coupled with electrospray ionization tandem mass spectrometry was developed to determine the contents of oxypaeoniflorin,albiflorin,paeoniflorin,benzoylpaooniflorin,baicalin,wogonoside,oroxylin A-7-O-glucuronide,baicalein,wogomn,and oroxylin A in Qingrehuoxue decoction,which has been used for the treatment of COPD.Separation was performed on a Poroshell 120-C18 reversed-phase column.The total run time was 18 min.Gradient elution was applied using a mobile phase of 0.1% formic acid in 5 mM ammonium/acetonitrile.The analytes were quantified on an Q-TOF mass spectrometer in electrospray ionization MS mode.It was found that the calibration curves for all analytes showed good linearity (R2>0.99) within the test ranges.The relative standard deviations (RSDs) for intra-and inter-day precisions were below 3.2% and 7.4%,respectively.The accuracy was evaluated by the recovery test within the range of 82.57% to 104.78% with RSD less than 5.0%.The sample was stable for at least 48 h at 4℃.The results showed that the new approach is accurate,sensitive,and reliable,and has been successfully quantified the 10 major constituents in three batches of Qingrehuoxue decoction.
