抗TLR2单克隆抗体在人THP1源性巨噬细胞中的抗炎作用

目的:在PMA诱导的人THP1源性巨噬细胞中检验抗TLR2单克隆抗体(anti-TLR2 mAb)的抗炎作用.方法:以200nM PMA诱导分化人THP1细胞72h,细胞完全贴壁后更换正常培养基培养5天,用巨噬细胞标志物ED1抗体经免疫荧光技术和流式细胞术鉴定分化结果后、将其分为三个实验组:control组、IgG组、anti-TLR2组,分别经PBS、IgG、anti-TLR2预处理1h后,加入Pam3csk4刺激24h收取细胞上清及总蛋白.最后用ELISA检测细胞上清中炎症因子IL-6、TNF-α的水平;用Western blotting检测MAPK和NF-κ B两条炎症信号通路的活化情况.结果:200nMPMA成功将THP1细胞诱导为单核巨噬细胞.与control及IgG组相比,anti-TLR2 mAb组细胞上清IL-6、TNF-α的水平显著降低;Western blotting结果也显示MAPK、NF-κB炎症信号通路的活化均有所降低.结论:Anti-TLR2 mAb在PMA成功诱导的人THP1源性细胞中减轻了Pam3csk4诱发的炎症反应.
