Transient receptor potential (TRP) channels are polymodal cation channels that provide one of the main pathways in excitable and non-excitable cells.Among them, the six thermo-sensitive TRP channels (TRPV1-4, A1, M8) are molecular sensors for environmental and internal temperatures required for survival and self-protection.These channels are nonselective cation channels which permeate Ca2+ on the plasma membrane.The Ca2+ influx through these channels may play major roles in their physiological functions, such as regulation of Ca2+ homeostasis and transmitter release/EC-coupling in variety of cells.Thus, it is important to determine the fractional contribution of Ca to the total ion influx, or fractional Ca current (Pf).In the present work, by combined measurements of patch-clamp and fura-2 fluorescence, we determined Pf through four thermo-sensitive TRP channels (TRPV1, TRPV2, TRPV3, TRPM8) in physiological solutions.From large to small, the fractional Ca2+ currents are in the order of 22% (TRPV2) > 16%(TRPV3) > 5.4% (TRPV1) > 4.7%(TRPM8).
