The interfacial interaction of sulfur oxidizing microorganisms (SOM) and sulfide minerals was characterized by using synchrotron radiation-based S/Fe/Cu K-edge X-ray absorption near-edge spectroscopy (XANES), X-ray Diffraction, hard X-ray microfocus, and soft X-ray spectromicroscopy together with other accessory approaches such as SEM/EDS, Raman spectroscopy, FT-IR spectroscopy,and electrochemical methods, in which the mineral surface S/Fe/Cu speciation, extracellular polymer substance (EPS) and SH group of microbial cells, adsorption of cells to the specific mineral surface sites were investigated during interaction between chalcopyrite or elemental sulfur and Acidithiobacillus ferrooxidans, Sulfobacillus thermosulfidooxidans, or Acidianus manzaensis.
