Migraine is a chronic disease that has complicated physiological, pathological and psychic reaction procedure.Present migraine animal models mainly focus on pathological changes in a certain stage during invasion process of migraine, fail to imitate complete pathological procedure of migraine, and have some deviations from clinical pathogenesy.Based on pathological changes in every stage of migraine morbidity, establishment and comprehensive, objective evaluation of animal models are important foundation for deep study on migraine nosogenesis, and its prevention.Theoretical foundation: migraine rat model with the feature of time phase is a new-type model on the basis of angiography theory.According to the feature of time phase of vascular changes during migraine morbidity process,dopamine, angiotonica, were administered to induce vasoconstriction of rat brains;after an interval, angised, vasodilator, were administered to provoke anapetia of rat brains, and dynamic procedure of pathological changes from migraine omen occurrence to every stages was well imitated, much alike to invasion process of this disease in clinic.This research emphasizes on observing ethological performance, vasoactive substances, neurotransmitter contents and brain haemodynamics of migraine model rats, approaching the evaluation method and index for initially established migrain models with the feature of time phase;then, compared study with angised migrain models generally applied was carried out to furtherly evaluate the reliability and adaptability of migraine rat models with the feature of time phase.Research evidences: first, ethological performances ofpiloerection, acceleration of blood flow rate in cephalic artery, and decrease of dopamine, norepinephrine, 5-hydroxytryptamine contents of our model indicated vasoconstriction stage.After the injection of angised to the model, nitrogen monoxidum generated in vivo precipitated the fierce expansion of blood vessel;the influence of nitrogen monoxidum on neurone function could result in delayed effect that will produce abnormal increase of c-fos gene expression in central nervous system;meanwhile, the raise of blood flow rate in cephalic artery and augmentation of 5-hydroxytryptamine contents suggested vasodilation stage.These two stages proved that the model performances and changes of pathological biochemistry induced by Dopamine and angised had some similarities with migraine morbidity of human.Second,compared with angised migraine models, the model with the feature of time phase was more reliable in behavior semeiology;these two models are similar in changes of vasoactive substances and neurotransmitters, furtherly confirming the reliability and availability of our model.The above conclusions initially manifested that the migraine rat model with the feature of time phase is the relatively stable and effective animal model for studying pathological and physiological mechanism of migraine, for developing prevention drugs and methods.
