Study on the Mechanism of Active Component in Radix Rehmanniae and Cornus officinalis for Regulating

  Object:Diabetes nephropathy (DN) is the typical diabetic complication.It includes glomerular mesangial cell proliferation,thickening of basement membrane and extracellular matrix increased.We will establish the injured model by Advanced Glycation End Products (AGEs) on glomerular mesangial cells (GMC).Furthermore,the effect of active component in Radix Rehrnanniae and Comus officinalis in DN by AGEs-RAGE pathyway will be studied.Method:AGEs levels were confirmed by detecting the levels of GMC appreciation rate,LDH.FN.COL-IV.After that,different concentrations (10.1.0.1pmol/L) of morroniside,loganin,oleanolic acid,ursolic acid,catalpol and verbascoside were added.After 24h,appreciation rate was detected,the cellular morphology and interior structure were observed.The RAGE,P38MAPK,ROS,FN,COL-IV and NF-κB were measured.Result:AGEs (200mg/L) could significantly induce the GMC proliferation (P<0.01) and LDH,FBL COL-IV secretion.Active components in Radix Rehmanniae and Cornus officinalis could significantly protect the GMC from AGEs (p<0.05,p<0.01),such as protect morphology,structure,reduce the degeneration degree of intracellular structure and organelle.Active components (1μmol/L) reduced the ROS,RAGE and P38MAPk significantly (p<0.05,p<0.01).Furthermore,the SOD and GSH were increased,the MDA,NF-κB,TGF-β,MCP-1,ICAM-1,LDH,LN,FN and COL-IVwere reduced Conclusion:The research indicated that the Active components in Radix Rehmanniae and Comus officinalis will protect the GMC by inhibiting oxidative stress and inflammatory reaction,regulating the extracellular matrix secretion.ursolic acid and catalpol can successfully restrain the RAGE activation.ursolic acid,loganin and verbascoside are better in response to oxidative stress.Morroniside and ursolic acid control the inflammatory response significantly.The catalpol and morroniside have positive e伍:ct in reducing the ECM and cell damage.As a result,the Active components in Radix Rehmanniae and Comus officinalis can prevention and cure the DN by regulate the AGEs-RAGE pathyway.
