In this study, an electrospray-based ionization source, a so called rotating eleetrospray ionization (RESI), was developed to interface high performance liquid chromatography (HPLC) with mass spectrometry to perform ultra ultra-high flow HPLC/Mass Spectrometry (MS) analysis.A RESI source is a plastic plate attached with an electrosprayer; the plate is connected to a step motor through a metal axis to move the plate in a wide circular motion.The sample solution contains a mixture of dye standards including methylene blue, acridine orange, malachite green and crystal violet, and was prepared for a reverse C18 HPLC analysis, with the ions being detected by an ion trap mass spectrometry.It was found that as the plate and electrosprayer rotated, large charged droplets effectively spun off the central line by centrifugal force, leaving small charged droplets and ions moving along the central line and entering the MS inlet.Hence, a HPLC combined with RESI/MS can be used for high-flow HPLC/MS analysis without the use of heated gas.The results suggest that the maximum HPLC flow rate for a HPLC-RESI system is 2 mL/min, without needing to divert the eluted sample solution.In addition, if a RESI source equipped with multi-electrosprayer and hot nebulizing gas is coupled to the HPLC system, this new apparatus can tolerate an even higher HPLC flow rate-5 mL/min.
