蟠桃褐腐病原菌生物学特性及致病相关酶活性的研究

  为了研究蟠桃褐腐病原菌不同环境条件下的生物学特性及细胞壁降解酶活性,我们利用PDA 平板培养法,PDB 摇瓶法对病原菌的菌丝生长、孢子萌发和芽管延长的最适温度、pH 光照条件进行了测定,同时利用DNS 法对细胞壁降解酶活性进行了测定,结果表明:温度会影响菌落形态,菌落生长的最适温度范围为20-28℃,当培养温度高于36℃时菌落不能正常生长;光照对菌落生长影响甚微;桃褐腐病菌孢子萌发和芽管延长的最适pH 为5.0,适宜生长环境为弱酸性,过酸环境其孢子萌发率明显降低;内切葡聚糖酶、β-葡萄糖苷酶、果胶甲基半乳糖醛酸酶、果胶多聚半乳糖醛酸酶酶活性均在28℃下最高;pH 为6.0 时内切葡聚糖酶、β-葡萄糖苷酶酶活性最高;PH7.0 时果胶甲基半乳糖醛酸酶、果胶多聚半乳糖醛酸酶活性最高;四种酶活性在培养5 天时的酶活性最高.
