Objectives Cardiovascular disease (CVD) , especially coronary heart disease(CHD), is one of the most dangerous diseases that seriously endanger the human health.A growing evidence from epidemiologic survey shows that periodontitis has a significant relation to CVD.However, the biological basis and possible mechanism of the correlation is not clear.Atherosclerosis (AS) is the basic pathological change of CVD, and infection and inflammation play important roles in the formation of AS.Epidemiological surveys found that C-reaction protein (CRP) has certain relevance to both periodontitis and CVD.This study aimed to study the function and possible mechanism of Porphyromonas gingivalis (P.g) and CRP in the correlation between periodontitis and CHD.Methods This study include following parts: 1.Analysis the correlation between periodontitis and CVD, and the CRP levels of peripheral blood in the periodontitis patients.2.Adhesion and invasion of P.g in endothelial cells; the effect of P.g and CRP on apoptosis of endothelial cells.3.The effect of P.g and CRP on chemotactic functions ofmonocytes.Results 1.There is a strong association between moderate or severe periodontitis and CHD that moderate or severe periodontitis were 2.5-fold more likely to have CHD; CRP was found to be significantly associated with an increased frequency of CHD.2.P.g has strong ability of adhesion and invasion in Human Umbilical Vein Endothelial Cells (HUVECs); P.g381 infected HUVEC demonstrated significantly increased adhesion of THP-1 cells; CRP and P.g can synergeticly increase the apoptosis of vascular endothelial ceils.3.P.g increased CC Chemotactic Receptor 2 (CCR2) expression on the surface of THP-1 cell in the early stage, enhancing the chemotactic capacity, thus contributing to the formation of AS plaque; CRP promoted the expression of CCR2 in THP-1 cells in a concentration-dependent manner and enhanced the chemotactic capacity of THP-1 cells.Conclusions P.g and CRP may be one of the biological bases that responsible for the association between periodontitis and CHD, and controlling the periodontal infection may decrease the incidence rate of CVD.
