To evaluate the chemical profile and anti-carcinogenic effect amongst totalsaponins fraction (TSF),25% ethanolic fraction (25EF),50% ethanolic fraction (50EF) and85% ethanolic fraction (85EF) by macroporous resin from leaves of Panax notoginseng.The simultaneous determination of 13 main sapoins,as well as full-scale chemical profileof different fractions,was analyzed by HPLC-DAD and LC-ESI-MSn.The anti-proliferativeeffects were determined on KP4 cells (human pancreatic cancer cells),NCI-H727cells(human lung cancer),HepG2 cells (human hepatocellular cancer) and SGC-7901 (humangastric adenocarcinoma) by cytotoxic assay.Chemical analysis indicated that 85EFpossessed the most abundant anti-tumor protopanaxdiol saponins,including markersaponins:F2,20 (R)-Rg3,20 (S)-Rg3 and Rh2.Besides,the results of MTT assay showedthat 85EF had the strongest anti-tumor effects among the four fractions.25EF showed theweakest anti-proliferative effects,while 50EF and TSF exhibited slight increasing inanti-proliferative activity.From the points of comprehensive utilization of resource,85EF,enriched with low polar PPD group saponins,is a new alternative source of anti-cancersaponins and a promising novel useful botanical product for cancer themoprevention.
