3T3-L1细胞诱导分化成熟阶段2种脂肪细胞因子的表达水平研究

本研究采用诱导分化培养法诱导小鼠3T3-L1前脂肪细胞分化为脂肪细胞.油红O染色法检测脂肪细胞的脂滴积累与脂肪细胞分化程度,发现在诱导分化后的7、8、9天能够观察到明显的脂质沉积,脂滴大小和数量均表现出逐渐增加的趋势.荧光定量PCR对增强子结合蛋白α(C/EBPα)和脂联素(ADIPOQ)两种脂肪细胞因子的基因相对表达量进行检测,结果发现在小鼠3T3-L1前脂肪细胞的分化过程中,随分化时间的延长,C/EBPα和ADIPOQ两个基因在3T3-L1前脂肪细胞的脂质高度累积阶段呈现出不同的表达量变化趋势.说明在小鼠3T3-L1前脂肪细胞分化为成熟脂肪细胞的过程中,C/EBPα对3T3-L1前脂肪细胞的分化和脂质积累可能起促进作用,而AdipoQ基因对3T3-L1细胞的诱导分化可能有促进作用,但与细胞内脂质的累积关系不大.
