甘蔗鞭黑穗病菌有性配合信号分子及相关基因的研究

  甘蔗鞭黑穗病是甘蔗的重要病害之一,病原菌Sporisorium scitaminea由不同性系(+、一)的单细胞菌体通过有性配合产生双核菌丝体侵染寄主。有性配合是甘蔗鞭黑穗菌生活史最重要环节之一,更是病原菌入侵寄主的必要前提。本项目以甘蔗鞭黑粉菌+、-两性系为材料,通过优化根癌农杆菌介导的(Agrobacterium tumefaciens-mediated transformation,ATMT)T-DNA随机插入方法,建立了一个一万五千个转化子的突变体库。转化子与野生型菌株的有性配合的筛选实验,获得了三个有性配合缺陷型突变体;通过hiTAIL-PCR和测序分析得到了突变体的T-DNA插入的位置及插入突变基因序列。进一步的实验将分析三个突变基因的功能,解析它们在甘蔗鞭黑粉菌有性配合过程中的作用。此外,课题通过前期筛选得到的生防菌绿色荧光假单胞杆菌YJHM58进行有性配合抑制实验,证实在黑粉菌的有性配合过程中存在与有性配合相关的可以扩散的小分子信号物质。正在优化进行的化学分离提纯的办法将对信号分子进行分离和结构鉴定。本研究从遗传和生化两个角度切入探索甘蔗鞭黑穗菌有性配合的通讯机制,旨在研发针对有性配合信号分子及传导通路为靶标的特异性的抑制剂和淬灭剂,建立一套准确有效的甘蔗黑穗病的生防技术。
