Capillary isotachophoresis (cITP) is a powerful separation tool,not only applied for highly efficient separation but also for focusing and enrichment of biomolecules over hundreds to thousands times.In general,the sample ions with intermediate electrophoretic mobility will be focused at the boundary of leading and terminating ions,which have the highest and lowest electrophoretic mobility in electrophoretic system,respectively.Here we demonstrate the moving and focusing of single DNA molecules and clarify the possible focusing mechanism.Based on the clarified cITP mechanism,we designed a strategy for focusing of DNA molecules and specific labeling of damaged DNA.With such a unique strategy,we expect to develop an approach detecting a single DNA damage using capillary electrophoresis-laser induced fluorescence imaging analysis.
