  Aim:Identification of tumor-associated gene in the process cell transformation with cell model Materials and Methods:JB6 cell line,which can transform by EGF treatment; suppression subtractive hybridization; Reverse dot blot; Northern blot; 5RACE et al.Results:a mouse Lon homology ATP-dependent protease (r-Lon),which exhibited 84% and 96% homology with human and rat respectively,was drastically up-regulated by EGF treatment.Both ATPase and serine protease domain,which are conservative in Lon,are all presented in this mouse counterpart.EGF-induced up-regulation of m-Lon is elicited by significantly increasing the transcriptional rate but not by mRNA stability and both ERK and PI3K pathways were essential for EGF-mediated m-Lon up-regulation.In addition,ErbB2 over-expressed human mammary epithelial cells express much higher level of Lon than normal mammary epithelial cells.
