Purpose:To study the effect of Cx43 (Connexin 43)and GJIC(Gap Junctional Intercellular Communication) by the MSC treatment in the prostate cancer DU145 cell.Methods:Cx43 mRNA and protein were examined by RT-PCR and WB,GJIC function by the SL/DT after 0,5,10,25μrnol MSC treatment.Further the growth of prostate cancer DU145 cell was tested by MTT and clone formation assay,and the BCL-2 protein level by WB after MSC treatment and Cx43 transfection;Results:5,10,25μmol MSC or exogenous transfection of Cx43 in DU145 cell can induce the Cx43 mRNA and protein,but not the GJIC function;Further MSC and up-regulation of Cx43.can reduce the BCL-2 level and inhibit the DU145 cell growth and proliferation (p<0.05);Conclusion:MSC can inhibit the DU145 cell growth and reduce the BCL-2 by the up regulation of Cx 43 which shows a new pathway of MSC in the anti-cancer effect.
