Epithelial-mesenchymal transition (EMT) is a process that converts epithelial phenotypes and behaviors to those of fibroblast-likemesenchymal cells during embryogenesis and cancer metastasis.During EMT, the cells lose cell-cell interaction, undergo an expression switch from marker proteins for epithelial to mesenchymal cells and gain increased cell motility and invasiveness.It is believed that EMT plays a critical role in initiating epithelial cancer metastasis including breast cancer by promoting the intravasation step of metastasis.Recent groundbreaking studies have demonstrated that mammary epithelial cells that have undergone EMT gain stem-like cell properties.Interestingly,other groundbreaking studies have recently shown that several Kruppel-like transcription factor [KLF] family members are critical regulators of the biology of embryonic stem cell or reprogrammed stem cells and EMT as well.The role of KLFs in the regulation of EMT, breast cancer stem cells, invasion and metastasis will be discussed with particular focus on KLF8.
