Astroglial TREK-1 Pathway Mediates lsoflurane-lnduced Memory Impairment in Middle-aged Mice

  Post-operative cognitive dysfunction(POCD)has been widely reported,especially in elder patients.An association betweenPOCDand inhalational anesthetics,such as isoflurane,has been suggested.The astroglial TWIK-related K+channel-l(TREK-1)controls several major cellular responses involved inmemory formation and is believed to take part in the formation of depression,cerebral ischemia and blood-brain barrier dysfunction.However,the specific role of astrocytic TREK-1 in mediating anesthesia-induced POCD remains unknown.In the current study,we determined that exposing mice to isoflurane affected astrocyte survival and memory and altered TREK-1s expression.In addition,TREK-1 0ver-expression exacerbated the anesthesia-induced memory impairment,while TREK-1 silence attenuated the isoflurane-induced memory deficits.We also observed that the deleterious effects of TREK-1 0n cognition involved the suppression of neurotrophic factors and glutamate transporters.Taken together,our data demonstrate that inhibition of astrocytic TREK-1 protects mice from cognitive impairment induced by anesthesia and that this protection occurs through regulation of the expression of neurotrophic factors and glutamate transporters.
