NRSF regulates its target genes via epigenetic mechanisms in PD cell model

　　Neuron-restrictive silencer factor/repressor element-1 silencing transcription factor (NRSF/REST) silences a large network of genes through histone deacetylation, methylation and other epigenetic mechanisms.Our previous study showed that the expression of NRSF target gene BDNF, TH, synapsin Ⅰ and UCH-L1 were transiently decreased in PD cell model.Here we investigated the effect of epigenetic modulation on the expression of NRSF target genes in the SH-SY5Y cells intoxicated with MPP+.CHIP assays showed that the binding of NRSF to the target gene promoters was increased.And we found the MPP+ treatment promoted deacetylation of histones H3, demethylation of H3-K4 and dimethylation of H3-K9 over the promoters of these target genes, which are the signatures of epigenetic gene silencing.Bisulfite sequencing results showed that the methylation pattern over the promoters of BDNF and TH transient altered in this PD cell model.
