Objective: To gain insight into the effects of amidation on the biological activity and mode of action in Cecropin XJ.Methods: Two custom peptides CecXJ-37C with a free C-terminus, and CecXJ-37N with an amidated C-terminus, were used to study the secondary structure, peptide stability, antibacterial and hemolytic activities, as well as the mechanisms of action.Results: Our study showed that amidation form was seen to increase efficacy approximately two to four folds when they were applied to different Gram-negative bacteria tested, and no obviously bactericidal difference (MIC < 1 μg/mL) was found when they were tested against Gram-positive bacteria (Table.1).
