目的:观察不同时间点注射抗氧化剂NAC后糖尿病小鼠心脏大小及功能变化,探讨其可能分子生物学机制,为临床糖尿病心脏改变治疗提供参考依据.方法:糖尿病鼠模型建立:采用STZ法建立I型糖尿病鼠模型,具体如下:C57BL/6小鼠,体重20-25g.腹腔注射STZ (Sigma-Aldrich,St.Louis,MO) 80mg/Kg.五天后,通过鼠尾静脉检测空腹血糖,血糖值超过13.5 mmol/L视为建模成功.NAC治疗及分组:STZ注射5天后,腹腔注射NAC (100mg/kg)进行治疗.根据注射NAC时间不同将糖尿病鼠随机分为5组,1周时开始注射为NAC1组,3周、5周及7周开始注射时分别为NAC3组,NAC5组和NAC7组,每组5只;另有5只糖尿病小鼠作为未治疗糖尿病组(UDM)及5只健康小鼠作为对照组.
