Research into the details and basis of the succession of plant establishment in the biochar augmented crop farming is of fundamental importance.This study has examined and documented the effects and influences on plant physiology of herbaceous biochar when used as a soil amendment, and with particular focus on what takes place below ground during the first 14 days of the plant growth.Multiple staining techniques (Ink and fluorochromes staining), and advanced 2D3D-imaging techniques (SEM, LSCM) were applied to analyse changes in: (1), root system growth and development (morphology and image analysis);(2), the density and spatial localisation of AMF hypae;(3), the abundance of the arbusculs and storage vesicles structures produced in the roots by arbuscular mycorrhizal fungal (AMF).Advanced DNA based molecular techniques, such as genera-specific PCR and PCR-DGGE were applied to determine the biodiversity indexes within the biochar augmented plant rhizosphere, compare to control plants and to monitor the AMF diversity shift in response to biochar addition over time.
