新型MIP-2γ/GM-CSF融合蛋白对DC的双重生物学功能及相关机制研究

  目的:制备新型MIP-2γ/GM-CSF细胞因子融合蛋白,研究其趋化和激活DC的双重生物学功能.方法:通过基因工程方法构建高表达MIP-2γ/GM-CSF融合蛋白的2种P.pastoris酵母工程菌株,并优化发酵、纯化条件,制备了高纯度的MIP-2g/hGM-CSF、MIP-2g/mGM-CSF融合蛋白.首先以Transwell体系检测融合蛋白对树突状细胞DC的诱导迁移效应.接着以流式细胞仪分析重组蛋白对DC表型的影响,检测其作用DC后引起的MLR反应和细胞因子分泌情况.进行动物体内实验,研究MIP-2γ/GM-CSF融合蛋白对CT26细胞转染的荷瘤小鼠的抗肿瘤效应,并研究了其相关的作用机制.
