Octodonta nipae(Maulik)is an important quarantine pest on palm in southern China and a candidate target for integrated pest management based on biological control.Thus investigations into its natural immune defenses are necessary.In the present study,the types of hemocytes were identified by transmission electron microscope(TEM),and phagocytosis was assayed by injecting both fluorescein isothiocyanate(FITC-)labeling Escherichia coli and Green Fluorescein Protein(GFP-)expressing E.coli into the pupae.Prophenoloxidase(PPO)-positive hemocytes were detected using an L-DOPA(dissolved in 35%ethanol)incubation method.We identified five hemocyte cell types: proemocytes(3.46%± 1.0%),plasmatocytes(44.10%± 3.3%),granulocytes(17.24%± 0.9%),spherulocytes(8.19%± 0.6%)and oenocytoids(15.21%± 1.5%).Granulocytes and plasmatocytes can phagocytize FITC-labelled E.coli and GFP-expressing E.coli in vivo 2-48h after injection.Plasmatocytes,granulocytes and oenocytoids showed PPO activity after incubating with L-DOPA.These data suggested that plasmatocytes and granulocytes were the main immune hemocytes in O.nipae.
