Ubiquitin is a highly conserved protein in eukaryotes, playing significant roles in maintaining protein homeostasis and cells signaling.Ubiquitin may be conjugated via a peptide bond between Gly-76 in the C-terminal and one lysine residue of the target protein.K48-linked ubiquitin dimer adopts a predominantly closed conformation, with a well-connected hydrophobic network at the inter-domain interface formed via the triplets, L8, 144, and V70,contributed from both monomers.In this work, we have investigated the ubiquitins dimerization landscape when the monomers are not covalently linked.
