目的:观察虎杖提取物对人外周血单个核细胞炎症因子基因的影响,以期阐明虎杖提取物抗痛风性关节炎的作用机制。方法:分离与培养人外周血单个核细胞,台盼蓝测细胞活力为95%以上,RT-PCR检测人外周血单个核细胞IL-1βmRNA、IL-8mRNA的表达。结果:虎杖提取物1000mg/L,100mg/L组IL-1βmRNA和IL-8 mRNA表达与空白对照组比较有显著性差异(P﹤0.01)。地塞米松磷酸钠组表达与空白对照组比较有显著性差异(P﹤0.01)。结论:虎杖提取物能抑制人外周血单核细胞炎症因子基因的表达,可能是其治疗急性痛风性关节炎的作用机制之一。
Objective: To observe the effect of Polygonum Cuspidatum extract on human peripheral blood mononuclear cells inflammatory factor gene in order to clarify the action mechanism of Polygonum cuspidatum extract against gouty arthritis. Methods: Peripheral blood mononuclear cells (PBMCs) were isolated and cultured. The cell viability was above 95%. The expression of IL-1βmRNA and IL-8 mRNA in human peripheral blood mononuclear cells were detected by RT-PCR. Results: The expression of IL-1βmRNA and IL-8 mRNA of Polygonum cuspidatum extract 1000mg / L, 100mg / L group were significantly different from the blank control group (P <0.01). Dexamethasone sodium phosphate group compared with the control group were significantly different (P <0.01). CONCLUSION: Polygonum Cuspidatum extract can inhibit the expression of inflammatory cytokines in human peripheral blood mononuclear cells, which may be one of its mechanisms in the treatment of acute gouty arthritis.
