钩吻素子对坐骨神经慢性束缚损伤模型大鼠脊髓3α-羟化类固醇脱氢酶表达的影响

  目的 观测在钩吻素子治疗CCI大鼠NPP过程中脊髓3α-HSD的表达动态变化规律.方法 :在大鼠CCI诱导的NPP模型上,于术后第3天开始,皮下注射给予钩吻素子(0.28,1.4,7 mg·kg-1)连续7d,以免疫组织化学方法,观测脊髓3α-HSD蛋白水平的动态变化;以Realtime-PCR法测定脊髓3α-HSD基因表达水平的动态变化.结果 免疫组织化学方法显示,3α-HSD在脊髓灰质均有分布;与sham组相比,CCI组、钩吻素子(7,0.28 mg· kg-1)大鼠脊髓背角3α-HSD蛋白表达随术后时间延长有上升趋势,其中钩吻素子(7 mg·kg-1)治疗组和CCI组相比,术后第9天脊髓3α-HSD蛋白表达的升高具有显著性差异,并且持续至第13天(停药后4d).Realtime-PCR显示,治疗组大鼠3α-HSD mRNA表达随术后时间延长逐渐增强趋势.CCI大鼠钩吻素子(7mg·kg-1)给药治疗后,术后第4天3α-HSD mRNA表达就有显著提高;术后第6天CCI组、钩吻素子(7,0.28 mg·kg-1)治疗组的3α-HSD mRNA表达均显著提高,但钩吻素子(7,0.28 mg·kg-1)治疗组与CCI组相比无显著差异;术后第9、13天,与Sham组相比,CCI组、钩吻素子(7,0.28 mg·kg-1)治疗组的3α-HSD mRNA表达均进一步显著提高,其中钩吻素子(7 nmg·kg-1)治疗组与CCI组、钩吻素子(0.28 mg·kg-1)治疗组相比均有显著差异.结论 钩吻素子抗NPP作用可能与上调脊髓3α-HSD酶表达有关.
