Heterogeneous expression and functional activity of ryanodine receptors in mesenteric artery smooth

  Objective: we aimed to investigate whether the expression and functional activity of RyRs would be different in mesenteric artery from patients with hypertension.Methods: RyR expression, [Ca2+]i were measured by real-time quantitative RT-PCR, immunofluorescence staining and wide-field fluorescence microscopy, respectively.Results: All 3 RyR subtype (RyR1 RyR2 and RyR3) mRNAs are expressed in HMASMCs, but their expression levels and distributions are different.RyR1 and RyR3 localize mostly to the central sarcoplasmic reticulum (SR),while RyR2 are especially the predominant isoform and distribute around the plasma membrane, colocalizing with BKCa channels.The expression level of RyR2 is significantly decreased in patients with hypertension.Activation of RyRs by caffeine (10 mM) or high K+ (60 mM) induces differential Ca2+ release.Moreover, the increases and decreases in [Ca2+]i induced by caffeine is faster in HMASMCs from patients without hypertension relative to those with hypertension.Conclusion: This study provides comprehensive evidence that RyRs are heterogeneous in expression and functional activity in HMASMCs from patients with hypertension, which may contribute to the diversity of excitation-contraction coupling and intimate communication between SR and sarcolemmal ion channel in different vascular smooth muscle cells.
