A strategy for simultaneously multiplexed DNA detection by an effective microfluidic droplets manipulation,which is called “assembly-line manipulation of droplets(ALMD)”,is presented in this article.Firstly,several droplets containing the same content are generated in the microchannel,and then fused with later generated droplets with desired content and size by the driving force of oil phase,respectively.Taking advantages of this effective method,the droplets fluorescence encoding can be achieved easily.Moreover,based on ALMD,the detection of different targets in droplets can be carried out simultaneously under identical conditions by concurrently imaging of droplets.Through the model of human immunodeficiency virus(HIV)gene sequence and variola virus(small pox,VV)gene sequence,it is proved that this method achieves simultaneous multiplexed DNA measurements with a significantly time-saving way and without different dye-labeled probes or complex operation procedures.In addition,it reveals the possibility of high-throughput biosensing with simple chip design and detection equipment.
