Betatrophin,a circulating hormone discovered in insulin-resistant states, has demonstrated possible physiological roles in the promotion of pancreatic β-cell proliferation [1] and the regulation of blood lipids[2-3].Based on bioinformatic analysis, several potential enzymatic-cleavage fragments were selected for construction and investigation.Peptides corresponding to the N-terminal region, middle region, and C-terminal regions of the betatrophin protein were prepared via solid phase peptide synthesis and antibodies were raised against them.Western blot analysis in human plasma found that the C-terminal betatrophin antibody was able to recognize both the 22 kD full-length betatrophin protein as well as two smaller betatrophin fragment peptides at 7.7 kD and 10 kD.
