Protein phosphatase-1 (PP 1) activity is involved in the regulation of a diverse range of cellular functions, acting to balance the actions of many protein kinases.Among the most important of these processes is that of cell division, where it acts to counterbalance the activities of mitotic kinases, notably Aurora kinase activity.Aurora kinase has multiple functions in the process of mitosis and cell division; it is frequently increased in tumors, and displays oncogenic properties.Little is currently known of the regulatory proteins that affect PP 1 activity during mammalian cell division.Our laboratory discovered a novel human PP1 inhibitor, Inhibitor-3 (Inh3), that inhibits PP1 activity with an IC50 in the nanomolar range.We have shown that Inh3 is a nuclear protein that is localized to the nucleolus together with the PP1 isoform in interphase cells by indirect immunofluorescence studies.
