Objective:To observe the effect of Radix Stemonae Concentrated Decoction on the lung tissue pathology and inflammatory mediators in COPD rats to explore its possible mechanism of action.Methods:SD rats were randomized into blank group,COPD model group and Radix Stemonae group by using random number method,10 cases in each of the 3 groups.Blank group:the animals were normally fed for 16 weeks.Blank group:the animals were normally fed for 16 weeks.Radix Stemonae group:the method was the same as the model group in the first 4 weeks,and from week 5 to week 16,apart from exposure to cigarette smoke,Radix Stemonae Concentrated Decoction was used for gavage of rats every day,3 times a day,1ml each time.Before the rats were sacrificed,lung function of the animals was tested.The lower right lung was removed for HE staining,pathological semi-quantitative judgment of the severity of lung inflammation,the severity of tracheal inflammation and the degree of alveolar fusion,and determination of the concentrations of TNF-α,IL-8 and LTB4 in BALF and serum by ELISA.Results:① Compared with the blank group,after exposure to cigarette smoke for 4 weeks,body weight of animals in the model group was decreased significantly(P<0.05);compared with the model group,after gavage therapy with Radix Stemonae Concentrated Decoction for 4 weeks(i.e.in week 8),body weight was increased(P<0.05).)② Compared with the blank group,Ri of rats in the model group was increased significantly(P<0.05),Cldyn was decreased significantly(P<0.05),and FEV0.2/FVC(%)and PEF were decreased significantly(P<0.05).Compared with the model group,airway Ri in the Radix Stemonae group was decreased significantly(P<0.05),Cldyn was increased significantly(P<0.05),and FEV0.2/FVC(%)and PEF were increased significantly(P<0.05).③ As for pathological changes in lungs,airway inflammation in the model group was aggravated,and significant emphysema occurred;in the treatment group,inflammation and emphysema also occurred,which were milder though.④ Compared with the blank control group,the concentrations of TNF-α,IL-8 and LTB4 in serum and BALF in the model group and the Radix Stemonae group were increased significantly(all P<0.05);compared with the model group,the levels of inflammatory mediators in serum and BALF in the Radix Stemonae group were decreased significantly(all P<0.05).Conclusion:Radix Stemonae Concentrated Decoction may mitigate and improve airway rebuilding in the lungs of COPD rats by inhibiting the release of inflammatory mediators,thus delaying the deterioration of lung function.)
