胃癌肿瘤干细胞的诱导及鉴定

  目的:探讨无血清悬浮培养法诱导胃癌肿瘤干细胞,并对其进行初步鉴定.方法: 无血清悬浮培养胃癌细胞系MGC803,观察肿瘤细胞球形成情况;流式细胞仪检测肿瘤细胞球中CD44+、CD54+、EpCAM及Trop2所占比例;检测肿瘤细胞球的细胞周期变化情况;定量PCR检测肿瘤细胞球中癌干调控基因Oct4、Snai1、Nanog及Trop2基因含量.结果:胃癌细胞系MGC803在无血清悬浮培养的条件下,大约14d左右能形成肿瘤细胞球且数量较多,肿瘤细胞球中CD44+、CD54+、EpCAM及Trop2含量明显高于正常培养条件下的MGC803细胞组(P<0.05);肿瘤细胞球中的细胞周期S期明显高于正常培养组(P<0.05);肿瘤细胞球中癌干调控基因Oct4、Snai1、Nanog及Trop2基因的表达明显高于正常培养组P<0.05).结论:胃癌细胞系MGC803在无血清悬浮培养条件能形成细胞球,这类细胞具有肿瘤干细胞的特征,出现高增殖状态,并能高表达癌干调控基因及促癌基因Trop2.
