铜离子作为氧化还原活跃的金属离子尽管在体内含量极低,但它是参与氧化代谢的金属酶的基本组分[1].癌细胞与正常细胞相比,需要更高水平的铜和活性氧来维持其恶性表型[2].此外,铜的水平直接和肿瘤的恶性程度相关[2].因此,癌细胞更加容易受损于铜离子介导的促氧化(促进活性氧生成)作用[2],支持着这一策略在选择性"杀死"癌细胞的重要性.作为我们促氧化介导的癌预防机制研究计划的部分[3-5],我们最近发现:(1)邻苯二酚结构是白藜芦醇及其羟基化类似物激活核转录因子Nrf2和诱导2相代谢酶表达及细胞抵御氧化性损伤的重要结构单元;其机制在于:它能够利用胞内的铜离子构建促氧化体系,产生邻醌.这种铜离子依赖的氧化转化对于Nrf2的激活是必须的.
