利用PCR方法扩增获得来自越南Mekong河和Dongnai河野生群体以及广东、广西、浙江养殖群体罗氏沼虾(Macrobrachium rosenbergii)线粒体DNA的COI基因部分序列进行测序,并从GenBank中下载了泰国和孟加拉国土著群体以及巴西引入群体的COI序列.通过序列分析我们共在150尾个体中发现了19个多态位点,定义了15个单倍型.五个群体中,两个野生群体的遗传多样性较为丰富(πDN=0.00286±0.00029,HdDN=0.694±0.059；πMK=0.00271±0.00033,HdMK=0.632±0.059).所有15个单倍型以及泰国和孟加拉国群体的序列进行系统发生分析的NJ树显示,所有序列分成了两个谱系：Mekong河和Dongnai河谱系(谱系1)以及泰国和孟加拉国谱系(谱系2),其中,来自浙江群体的Hap14和Hap15属于谱系2,其余个体均来自谱系1.五个群体的遗传距离分析显示,广东养殖群体和广西养殖群体的遗传关系最近(DxyGD-GD=0.000945),浙江养殖群体与两个野生群体的遗传距离最远(DxyMK-ZJ=0.004679；DxyDN-ZJ=0.004610).综上,我们认为广东和广西养殖群体原种来源于Mekong河或者Dongnai河；而浙江群体则分别来自于Mekong河或Dongnai河和泰国或孟加拉国.
