Protective effect of Emodin Lipid Nano-microbubble on ventilator induced lung injury in mice

  Objective Mechanical ventilation (MV) has become an important clinical treatment of acute and severe patients, however ventilator induced lung injury (VILI) is one of the most serious complications of mechanical ventilation.The main pathophysiological manifestations of lung injury induced by mechanical ventilation are inflammatory reaction and inflammatory cell infiltration.Studies show that emodin has anti-inflammatory effect and antioxidant effect, this project is designed to compare the protective effect of the different kind of dosage form of emodin on VILI.Methods The model of acute lung injury was induced by mechanical ventilation in mice which were randomly divided into 4 groups: normal control group (Con), VILI group (VILI),emodin treatment group (Emodin Inj.), emodin bubble treatment group (emodin bubble).Except the normal group, the other three groups undergo on large tidal volume mechanical ventilation (tidal volume vt 42ml/kg, respiratory frequency 40 beats / min, PEEP=0) to form mechanical model of lung injury.Two groups were injected respectively with emodin injection and emodin lipid nano-micro-bubble, ten minutes later, the group of lipid nano-micro-bubble group is given ultrasound for 60 seconds on lung.These methods including EMSA, Western-Blot ,ELISA and HE stained for histopathological examination were used to observe the expressions of NF-κB and IκB-α, to detect the levels of inflammatory mediators of TNF-α, IL-1β, IL-6, and to assess the pathological changes Results compared with the normal group, the expression of the model group was high, which showed that the model was successful.Meanwhile, Emodin injection and emodin lipid micro bubbles can reduce the lung injury induced by mechanical ventilation.The concentrations of IL-6, IL-1, MIP-2 and TNF-a in bronchoalveolar lavage fluid were significantly reduce and the ratio of wet and dry change.In addition, according to the pathological morphology, the effect of emodin bubble on lung injury was also obvious.Conclusion This experiment shows that the mechanical model is successful.Emodin has the protective effects of antioxidant and lung injury, Lipid vesicles were better, it was speculated that the size of the nanobubble was easy to gather in the lungs, and the ultrasound was helpful for the release of the drugs in the lungs.The results show that emodin can reduce the inflammatory response of lung injury, and protect the lung injury induced by mechanical ventilation.
