Epithelial-mesenchymal transition is the key process driving cancer metastasis.Oncogene/self renewal factor BMI-1 has been shown to induce EMT in cancer cells.Recent studies have implied that noncoding microRNAs (miRNAs)act as crucial modulators for epithelial-mesenchymal transition.The aims of this study was to determine the roles of BMI-1 in inducing epithelial-to-mesenchymal transition of endometrial cancer cells and the possible role of miRNA in controlling BMI-1 expression.We demonstrated that the expression of an important oncogene BMI-1 was suppressed by miR-194 at the post-transcriptional level in endometrial cancer.The suppression of BMI-1 by miR-194 is essential in regulating epithelial-mesenchymal transition of endometrial cancer cells.The expression of miR-194 was significantly reduced in endometrial cancer tissues and patients with higher expression levels of miR-194 survived longer.These results provide a critical foundation for developing future therapeutic strategy to effectively suppress human cancer metastasis by targeting BMI-1 using miR-194 or small molecule mimics.
