In order to explore the extraction methods of genomic DNA and reveal the genetic differentiation among geographic populations of Leguminivora glycinivorella(Matsumura),the CTAB method,the SDS method and a commercial extraction kit method were used to extract the genomic DNA and a part of the mitochondrial cytochrome c oxidase subunit Ⅰ(COⅠ)gene was sequenced from 7 geographical populations.The sequence variability of the COⅠ gene and genetic differentiation among them were analyzed,and then the molecular phylogenetic tree was constructed by the neighbor-joining method.⑴ On the basis of protein and nucleic acid radiometry,the purity and quality of genomic DNA extracted by the commercial kit were higher than those by the other two methods.⑵ The NJ tree suggested that the haplotypes were clustered into three clades associated with geographic regions.It showed that there was a close relationship among Helong,Jiamusi,Changchun,Cangzhou and Shenyang populations in first branch,both the Baiyin and Yan′an populations formed separate branches.It seemed that the differentiatin of the 7 populations of Leguminivora glycinivorella was realted to their geographic distance.
