  When infected by the Vibrio splendidus,the sea cucumber Apostichopus japonicus invokes an innate immune response that protects the species from pathogenic attack.MicroRNAs have emerged as key players in this host pathogen interactions.In the current study,we identified two important microRNAs,miR-137 and miR-2008,and its downstream-target gene of betaine-homocysteine S-methyltransferase(BHMT)combined by RNA-seq.Using a dual-luciferase reporter fused to the 3UTR of BHMT,we found a 61.59%(P=0.0002)and 36.60%(P=0.0359)reduction in luciferase activity in HEK293 cells compared to controls for miR-137 and miR-2008,respectively.Transfection of miR-137 mimics in sea cucumber coelomocytes significantly decreased,whereas miR-137 inhibitors increased,the mRNA and protein expression levels of BHMT.Meantime,homocysteine contents were also altered accordingly.More importantly,miR-2008 mimics and inhibitor showed no effect on mRNA levels of BHMT,but the concentration of BHMT protein displayed significant change.Our present study firstly confirmed that the differential roles of miR-137 and miR-2008 in non-model invertebrates pathogenesis.
