Gene therapy is an attractive approach for the treatment of cancer because of its ability to localize toxicity to tumor cells.Following delivery of the suicide gene to target cells, an inactive prodrug is administered.The prodrug is then converted to a cytotoxic compound through the actions of the suicide gene product thereby destroying cancer cells.One such gene product, cytosine deaminase (CD), is responsible for deaminating cytosine to uracil as well as the cytosine analog 5-fluorocytosine (5FC) to 5-fluorouracil (5FU), a highly potent anti-cancer agent.Because the conversion of 5FC to 5FU by cytosine deaminase is rate limiting, high doses of 5FC are required and result in non-specific side effects that restrict administration at therapeutically effective doses.
