Real-time quantitative PCR (qPCR) was used to enumerate seven unique diazotroph phylotypes in seawater samples collected during four cruises to the North Atlantic Ocean.TaqMan? probes (Applied Biosystems) and primers were designed to target the nifH gene which encodes the iron-protein subunit of nitrogenase,the prokaryotic enzyme responsible for nitrogen fixation.A total of 142 samples,covering a sampling area of 0°N to 42°N and 67°W to 13°W,were analyzed.The cyanobacterial phylotypes dominated in abundance (92.9% of all nifH copies detected) and were the most widely distributed.Among the cyanobacterial phylotypes,the filamentous type,which included both Trichodesmium and Katagnymene nifH,were the most abundant (51%),followed by Group A,an uncultured unicellular cyanobacterium (32.7 %),and Gam A,an uncultured γ –proteobacterium (5.7%).
