目的:糖尿病肾病(Diabetic nephropathy,DN)是糖尿病(Diabetes Mellitus,DM)全身性微血管合并症之一,也是DN患者主要的死亡原因.本实验拟建立糖尿病肾病动物模型,观察芪药消渴胶囊对复合法致实验性糖尿病肾病(DN)大鼠的保护作用.实验方法: SD大鼠(雄性,体重200±20g,由第四军医大学实验动物中心提供,合格证号SCXK(军)2007-007.饲养环境:陕西中医学院SPF动物饲养观察室)用高糖高脂饲料喂养合并单侧肾切除,配合一次性腹腔注射40mg/kg链脲佐菌素制造糖尿病肾病模型,造模成功后,大鼠眼眶后静脉取血,检测血肌酐(SCr)、尿素氮(BUN).芪药消渴胶囊组2.4、1.2、0.6g/kg灌胃给药治疗12W后,颈总动脉取血,检测SCr、BUN、超氧化物歧化酶(SOD)和丙二醛(MDA),并在给药4、8、12 W时收集尿液,检测24 h尿微量白蛋白的变化.
